EFFECTS OF LONG-CHAIN NONCODING EPIDERMAL GROWTH FACTOR RECEPTOR ANTISENSE RNA 1 ON THE PROLIFERATION AND MIGRATION OF RENAL CANCER CELLS

摘要:
Objective: We sought to analyze the effects of long-chain noncoding epidermal growth factor receptor antisense RNA 1 (EG-FR-AS1) on the proliferation and migration of renal cancer cells. Methods: The effect of EGFR-AS1 on the proliferation of renal cancer cells was detected by Colony Counting Kit 8 assay, the effect of EGFR-AS1 on the migration ability of renal cancer cells was detected by scratch test, the effects of EGFR-AS1 on cell cycle and apoptosis were analyzed, and the changes in EGFR messenger RNA (EGFRmRNA) expression after interfering with EGFR-AS1 expression were detected by real-time quantitative polymerase chain reaction (PCR). The western blot method was employed to detect changes in EGFR protein expression after interfering with EGFR-AS1 expression. Results: When compared with in the control group, the proliferation of renal cancer cells decreased significantly following knockdown of EGFR-AS1, which indicated that EGFR-AS1 could promote the proliferation of renal cancer cells. The scratch test results additionally showed that, as compared with the control group, the migration ability of the renal cell carcinoma cell line downregulated by EGFR-AS1 was significantly, decreased. Flow cytometry suggested that the expression of EGFR-AS1 was blocked in the G2/M phase of renal cancer cells after interference, and the expression of EGFR-AS1 was significantly higher than that in the control group (P<0.05). The results of RT-PCR showed that the expression of EGFRmRNA decreased significantly after interference by EGFR-AS1 when compared with in the control group. The results of western blot assay suggested that EGFR protein expression was decreased significantly after the expression of EGFR-AS1 was interfered with. Conclusion: Long-chain noncoding EGFR-AS1 is highly expressed in renal cell carcinoma, which can promote the proliferation and metastasis of renal cell carcinoma. The mechanism may be related to the maintenance of EGFRmRNA stability by EGFR-AS1, and this information can be used as a new marker for the clinical treatment of renal cell carcinoma.
